Mutagenesis is the basis for functional genes identification and germplasm innovation in crops. In this chapter, besides natural mutations as an important way to obtain mutant plants in cotton, the research progress in cotton mutagenesis strategies is reviewed here. Construction of relevant mutant libraries has been applied through physical or chemical mutagenesis. Artificial mutagenesis includes T-DNA insertion and transposable mutation, as well as development and optimization of precise mutagenesis approaches, such as a series of advanced CRISPR/Cas9 systems. With the development and improvement of modern mutagenesis technologies, elite cotton mutants can be created and selected to provide important germplasm resources for subsequent gene function investigations, germplasm innovation, and the breeding for new varieties of cotton.
