Introduction to Cryo-TEM and Related Sample Preparation

Cryo-TEM is a powerful and innovative imaging technique deals with preservation of sample structure at cryogenic conditions, during imaging. The basic principle, working with unique features related to respective techniques have been discussed in detail. Further, the importance and preference of cryo-TEM w.r.t. conventional TEM during specimen analysis also has been explained. Broadly w.r.t. specimen analysis under cryo-TEM, we have segregated respective samples into biological and soft specimen categories. Summary of key factors during cryo-TEM analysis viz. vitrification, slow or rapid freezing conditions, grid handling, artefacts generation, imaging precision w.r.t. innovative results and respective applications via emerging technologies has been presented.
