The β2-adrenergic receptor (β2AR) serves as a model for understanding how G protein-coupled receptors (GPCRs) administrate intracellular signaling. As a crucial element of the adrenergic signaling cascade, β2AR plays an essential role in mediating catecholamines in various tissues and organs. The receptor undergoes agonist-mediated endocytosis, a process that relies on arrestins, which bind to activated GPCRs and are vital for their function. So, an intricate understanding of the mechanisms underlying endocytosis and subsequent signaling events is crucial to explain the complex cellular response network to adrenergic stimulation. This chapter contributes to a brief overview of the complex signaling pathways involved in β2-adrenergic receptor endocytosis, the growing significance of endocytic trafficking, and the downstream signaling cascades triggered by these receptors, focusing on the internalization and signal transduction processes.
