Meningioma Development: Meningeal Embryology and Tumorigenesis

The anatomy of the human meninges is complex both at the macroscopic and the microscopic level. The histological subtypes of meningiomas are equally varied, raising questions regarding the cell of origin of meningiomas. Molecular classifications of meningiomas reveal a clear relationship between NF2-associated meningiomas of the convexity and the neural crest origin of the meninges, whilst TRAF7-associated and Hedgehog signalling meningiomas develop in the skull base where evidence suggests this is derived from mesoderm. We hypothesise that further understanding of human meningeal embryology will reveal mutually exclusive cells of origin for genotypically distinct meningioma necessitating distinct translational pipelines for targeted therapies in instances where patients are unable to be treated with surgery or radiotherapy.
