Sero-epidemiology uses measures of antibodies against a pathogen within a population to understand the transmission and epidemiology of infectious diseases. As antibodies persist for longer in the immune system than pathogens, serology offers an insight into exposure over a wider time frame than traditional diagnostics which capture active infections. This is useful as it allows for the reconstruction of transmission history from a representative cross-sectional survey over a period of time. This chapter outlines the core concepts of sero-epidemiology and its applications, with a focus on malaria. The principles in serological methods and study design, molecular, epidemiological and statistical considerations, and the limitations of sero-epidemiology as a tool are discussed, with examples of real-world applications.
