Diagnosis and Management of Diffuse Large B-Cell Lymphoma

Diagnosis and management of diffuse large b-cell lymphoma (DLBCL) is challenging. Recent genomic advances allow discrimination of aggressive lymphoproliferative entities for cytoreductive intensification and cellular therapy adoption. Han’s algorithm subclassifies the disease according to cell of origin (COO) in Germinal Center B-cell-like (GCB) with CD10 and BCL6 expression or Activated B-Cell-like (ABC) with overexpression of nuclear factor Kappa-β (NF-kβ) and BCL-2. However, genomic heterogeneity is high, suggesting that a better understanding of molecular classification is pivotal for therapeutic efficacy. In this chapter, we describe immunophenotypic, cytogenetic, and molecular features of DLBCL to illustrate current standard management.
