Covalent Inhibitors of MAP Kinases

Several covalent inhibitors such as aspirin, Penicillin G, and Omeperazole have been used for decades. Covalent inhibitors form a covalent bond with a specific target protein and effectively disrupt the activity of the target protein, which involve disease processes such as cancer progression, inflammatory responses, or infectious diseases. The reaction mechanisms of some FDA-approved covalent inhibitors are depicted. The binding mode of covalent inhibitors of MAP kinases is summarized.
