Non-ATP Site Inhibitors and Bi-dentate Inhibitors

The ATP site of the MAP kinases shares a common amino acid sequence. The ATP-competitive inhibitors are less selective and cause undesirable toxicities. The non-ATP site inhibitors are a class of compounds that inhibit kinase activity by binding to sites other than the ATP-binding pocket. The peptides and small molecule inhibitors are described here. Bi-dentate MAP kinase inhibitors are a specific class of compound that binds to both ATP site and allosteric site.
