Immunophenotypic characterization by flow cytometry (FCM) is an essential tool in the diagnostic workup of acute leukemias. Primarily, immunophenotyping at diagnosis is used for lineage assignment, which is essential for treatment choice. It also allows for maturational subclassification and antigenic profiling which informs in many cases about underlying genetic lesions. In addition, antigenic marker profiling is a prerequisite for minimal measurable disease assessment during follow-up, which is an essential tool for treatment stratification, as well as for selection of appropriate targeted immunotherapies.
