This chapter provides a comprehensive overview of advanced techniques in diffusion tensor imaging (DTI) and fiber tractography for clinical research and practice. Beginning with an introduction to DTI, it outlines the specific scanner requirements and data acquisition protocols necessary for accurate imaging. The process of preprocessing raw data is then discussed, followed by standards of fiber tracking methodologies. An extensive review of white matter tract anatomy, including association, commissural, projection, and other fibers, elucidates their distinct characteristics and clinical significance. Practical guidance is provided on defining seed points for fiber tracking, with specific examples focusing on major white matter tracts. The chapter concludes with a discussion on the clinical applications and limitations of tractography, including case examples.
