Investigation of Antioxidant, Anti-inflammatory and DNA Cleavage Activities of Ruthenium(II)-Bipyridine-Triazole Complex

The antioxidant, anti-inflammatory and DNA cleavage ability on plasmic DNA of [Ru(dtbpy)2(pytrzSH)2]2+ complex (dtbpy = di-tertiary-butyl-2,2′-bipyridine and pytrzSH = 5-(3-pyridyl)-4H-1,2,4-triazole-3-thiol) complex has been investigated. The [Ru(dtbpy)2(pytrzSH)2]2+ complex shows absorptions at 285 and 470 nm. The FT-IR spectrum of the [Ru(dtbpy)2(pytrzSH)2]2+ complex displays absorption bands in the region 3426–664 cm−1. The antioxidant potency of the complex is carried out using the DPPH assay method. The IC50 value of [Ru(dtbpy)2(pytrzSH)2]2+ complex is 523.80 μg/mL. The anti-inflammatory potency of the complex is determined by the protein denaturation inhibition assay method. The IC50 value of [Ru(dtbpy)2(pytrzSH)2]2+ complex is 245.44 μg/mL. The [Ru(dtbpy)2(pytrzSH)2]2+ complex has the ability to cleave the plasmid DNA at various concentrations ranging from 25 to 100 μg/mL. The treated plasmid DNA produce a smear indicating DNA damage, and the cleavage is observed in lanes 3, 4, and 5 of the treated plasmid DNA. Thus, the biological evaluation of the synthesised complex highlights a promising trajectory for future investigations in clinical diagnosis and therapeutic interventions.
