Strategic Defenders: Antimicrobial Peptides and Their Impact on the Innate Immunity

The surge in life-threatening infectious diseases, augmented by the escalating challenge of antibiotic-resistant strains, has prompted a critical need for effective treatments on a global scale. Antimicrobial peptides (AMPs) are versatile molecules with a wide-ranging ability to directly combat bacteria, yeasts, fungi, protozoa, viruses, and even cancer cells. Expressed in diverse cells such as epithelial cells, phagocytes, neutrophils, lymphocytes, and keratinocytes, these peptides are recognized for their pivotal contribution to immune response through a variety of mechanisms, including chemotaxis, modulation of host cell responsiveness to TLR ligands, stimulation of angiogenesis, enhancement of leukocyte/monocyte activation and differentiation, and modulation of the expression of pro-inflammatory cytokines/chemokines, underscoring their widespread significance and utility. These gene-encoded molecules, typically short and amphipathic, exhibit broad-spectrum antimicrobial activity. Despite hundreds of millions of years of coevolution with microbes, AMPs have retained their efficacy in combating infectious agents, offering a promising pathway for the development of a novel class of antimicrobial molecules, including numerous antibiotics. Beyond serving as a cornerstone in innate host defense against microbial colonization, AMPs act as a crucial link connecting the innate and adaptive immune responses, laying the groundwork for advances in the development of innovative therapeutic agents. The distinctive and varied functions of AMPs have garnered significant interest in understanding the fundamental biology of the innate immune system and utilizing these small molecules as a framework for the development of novel anti-infective drugs. This comprehensive review explores the intricate interplay between AMPs and innate responses. We examine the multifaceted roles of AMPs, ranging from their direct antimicrobial actions to their modulation of immune cell activities. A focal point of this chapter is the exploration of AMPs, essential elements in the innate immune arsenal. The discussion encompasses the definition and classification of AMPs, unraveling their structural diversity and functional classifications. A deeper dive into the intricate structure and mechanisms of AMPs provides a nuanced understanding of their multifaceted modes of action against a spectrum of pathogens. The narrative extends to the profound role of AMPs in host defense, emphasizing their contributions to immune modulation and the elimination of invading pathogens. Furthermore, we discuss intricate interactions between AMPs and other innate immune molecules, highlighting the synergistic efforts that fortify the host’s defense mechanisms. This chapter advances our comprehension of the native defense mechanism. It also highlights the important role of AMPs as conductors in orchestrating the overall defense.
