Ameliorative Effects of Oleoyl Chalcones on Liver and Cardiac Tissues Oxidative Damage and Hypertrophy Induced by High Fat Diet in Rats

High-fat diet (HFD)-induced obesity could lead to cardiac dysfunction, NASH, and liver fibrosis. Oleoyl chalcones is a common nontoxic polyphenol having a broad range of pharmacological and biological activities. The present study aimed at evaluating the anti-oxidative and histological effects of oleoyl chalcones on HFD-induced liver and cardiac dysfunctions. Fifty adult male SD rats were used and divided into five groups: Normal control (LFD): rats fed with a low fat diet, Positive control (Obese): rats treated with high-fat diet-fed (HFD), Orlistat: high-fat diet fed rats treated with Tetrahydrolipstatin (Orlistat), 5b and 5c groups: high-fat diet-fed rats treated with oleoyl chalcones (5b and 5c), respectively. By assessing the amounts of reduced glutathione (GSH), peroxidase glutathione (GPx), catalase (CAT), superoxide dismutase (SOD), and malondialdehyde (MDA), oxidative stress and antioxidant parameters were assessed in liver and heart tissues. Moreover, histopathological investigations of liver and heart tissues were examined. Results indicated that treatment with oleoyl chalcones (5b and 5c) restored the oxidative disturbance through significant increase of GSH, GPx, CAT and SOD with significant decrease in MDA levels in liver and cardiac tissues. Furthermore, amelioration of tissues dexterous effects induced by HFD of liver and cardiac was noted histopahologically. We concluded that oleoyl chalcones 5b and 5c showed an effective hepatic and cardiac protective agent against obesity-induced cardiac and NASH hypertrophy.
