Nuclear Pore Complex-Regulated Transcription and Memory

The nuclear pore complex (NPC) stands as a marvel of cellular architecture that orchestrates the bidirectional traffic of macromolecules between the nucleus and the cytoplasm. The NPC also physically interacts with chromatin to regulate genomic organization, transcriptional regulation, and epigenetic modulation. Aberrations to the NPC and its components have diverse effects and can lead to human diseases. Loss of certain nucleoporins and transcription factors that interact with the NPC disrupt the interaction with the genome, leading to transcriptional and epigenetic defects. While certain genes require physical interaction with the NPC for full transcription, others require physical interaction with the NPC for proper silencing. In this chapter, we will discuss the role of the NPC in regulating gene expression and the three-dimensional architecture of the genome.
