Marker Selection of Russian Rice Varieties Resistant to Prolonged Flooding

Rice resistance to prolonged flooding is necessary for competing with weeds when using herbicide-free technology. The creation of such rice varieties is an urgent problem, as it will reduce the cost of cultivation and reduce the impact of pesticides on the ecosystem. The article presents the results of the study of 180 breeding rice samples by methods of molecular DNA labeling based on the presence of the Sub1A flood resistance gene. In the course of the study, methods were used: homogenisation and isolation of DNA, polymerase chain reaction with specific molecular markers Sub1A203 and Sub1A-1 AEX, horisontal gel electrophoresis on 2% agarose gels, visualisation in ultraviolet light, analysis of electrophoregrams. As a result of studies in 28 rice samples, the locus Sub1A was identified (3121 (Inbara3 × Innovator), 3122 (Inbara3 × Innovator), 3123 (Inbara3 × Innovator), 3124 (Inbara3 × Innovator), 3125 (Inbara3 × Innovator), etc.); the Sub1A-1 allele associated with resistance to deep flooding was identified in 36 samples (354 (Inbara3 × Innovator), 1177 ((Inbara-3 (Sub-1) × Innovator) × Contact), 1178 ((Inbara-3 (Sub-1) × Innovator) × Contact), 2158 (Contact × Kharsu80A), 3131 (Inbara3 × Innovator)). Eleven samples were identified by two marker systems as having the Sub1A locus and the Sub1A allele-1 (1177, 1178, 3131, 3132, 3675, 988, 3121, 3123, 5578/2, 5618/2 and 5618/4) Selected promising lines for breeding work.
