Nanoluciferase (NanoLuc®) is a small, bright reporter enzyme known for its versatility. Detection of nanoluciferase, dependent on the substrate furimazine, is highly sensitive and can be adapted to multiple cellular assays. This chapter describes the use of codon-optimized nanoluciferase as a reporter for promoter activity, to assess protein topology and to investigate protein–protein interactions in Clostridioides difficile.
