Homemade Locus-Specific FISH Probes: Bacterial Artificial Chromosomes

In addition to the well-known applications of bacterial artificial chromosomes (BACs) in classical molecular genetics, these probes are also well suited for molecular cytogenetic studies. BACs are currently the most commonly used locus-specific probes in FISH. Various applications are possible, such as gene mapping, FISH banding, determination of chromosome breakpoints, characterization of derived chromosomes, investigations of interphase architecture, or karyotype evolution studies. Here, the basic principle of how BACs can be hybridized in situ on chromosome preparations is described. In addition, an overview of possible questions that can be investigated with BACs as FISH probes is provided.
