Fluorescence In Situ Hybridization for Plants: Methods of Chromosomal Preparation

Fluorescence in situ hybridization (FISH) is one of the most powerful and widely used methods in plant cytogenetics. The results of FISH depend primarily on the quality of the chromosome preparations, which in turn is determined by the appropriate pretreatment of the materials and the technique used to obtain the chromosome preparations. Plant species differ considerably in their genome size, chromosome number and length, cell cycle duration, and many other characteristics. For this reason, numerous different protocols have been developed for the preparation of mitotic and meiotic chromosomes from plants. Here, we present protocols for the pretreatment of meristematic tissue from plants with small, medium, and large chromosomes, as well as various methods of chromosome preparation for FISH.
