It is well known that chromosome in situ hybridization enables the unambiguous identification of targeted human somatic chromosomes. Over the years, various fluorescence in situ hybridization (FISH) techniques have been developed, and following mitosis studies, these various techniques have also been used to perform meiosis analyses. The application of FISH protocols to meiotic cells requires adaptation of standard protocols to the specific characteristics of these cells. Special sample fixation is usually required, and in some cases, samples must undergo special pretreatment to ensure successful FISH. The application of FISH in meiotic cytogenetics has proven to be very useful and has provided us with important information about many of the processes that occur during human gametogenesis. The protocols described in this chapter illustrate the processing of different meiotic samples and the FISH procedures.
