Molecular cytogenetic approaches applying smaller, locus-specific probes such as cDNA, plasmids, cosmids, fosmids, P1-clones, bacterial artificial chromosomes (BACs), or yeast artificial chromosomes (YACs) can sometimes be compromised by inefficient hybridization. In addition, FISH results may be required within a few hours, particularly in diagnostics. Here, we present a FISH protocol using microwave treatment that results in better hybridization efficiency with smaller probes and evaluable results within a few hours.
