Molecular cytogenetics originally comprised two basic approaches: fluorescence in situ hybridization (FISH) and primary in situ hybridization (PRINS). Today, FISH is the only routine approach still used in research and as molecular cytogenetics. This section describes the basic protocol for performing FISH using commercial and/or self-made DNA probes. A protocol for FISH on tissue sections can be found in the Chap. 23 . In addition to directly labeled probes, indirectly labeled probes can also be used; both variants are described here.
