RNA Extraction Strategy for Algae Rich in Polysaccharides and Polyphenols

RNA extraction in algal research is particularly challenging due to the high levels of polysaccharides and polyphenols that interfere with the purification process. This chapter presents a streamlined RNA extraction strategy, covering all steps from sample preparation to final purification. The method utilizes a CTAB-based lysis buffer and targeted separation steps to effectively eliminate proteins, polysaccharides, and polyphenols, employing lithium chloride for RNA precipitation. Designed for broad applicability across algal species, this strategy can be easily adjusted and performed with basic laboratory equipment, providing an accessible and effective tool for algae research.
