MicroRNAs (miRNAs) are key post-transcriptional regulators of gene expression. Accurate quantification of miRNAs is essential for understanding their biological functions. In this chapter, we first describe methods for extracting miRNAs from cells, tissues, and body fluids. We then outline the principles and procedures for reverse transcription and quantification using RT-qPCR and digital droplet PCR (ddPCR).
