Extraction and Measurement of Ergosterol in Mushroom and Mushroom Substrate

Several distinct lipids may serve as useful indicators for differentiating between species of fungus. As a subclass of lipids, ergosterol is a particular kind of sterol. It is mostly present in fungal cell membranes. However, fungi differ in their cell size, shape, cell wall structure, and other attributes, so it is necessary to investigate extraction techniques to recover these bioactives effectively. The most widely utilized of these methods is the quantification of ergosterol, a biomarker that is mostly exclusive to fungi. The ergosterol from fungal-colonized substrates is extracted using methanol and measured by high-performance liquid chromatography (HPLC) with UV detection at 282 nm. This chapter designs a method to quantify the mycelia in fully colonized mushroom substrate, such as spent mushroom substrate (SMS).
