Flow Cytometry Analysis of IFNγ-Induced PD-L1 Cell Surface Expression in Ovarian Cancer Cells

ImmunotherapiesImmunotherapies blocking immune checkpoint PD-1/PD-L1 signaling have shown great promise in a variety of cancers, but most patients fail to respond or develop resistance. One of the factors determining the responsiveness to PD-1/PD-L1 targeting immunotherapies is surface expression of PD-L1 on cancer cells. Here we describe a flow cytometry protocol for the analysis of cell surface expression of PD-L1 induced by IFNγ in ovarian cancer cells, by using antibody that specifically recognizes the extracellular domain of PD-L1.
