Ultrastructure Expansion Microscopy for the Analysis of Trypanosoma cruzi Infection

We describe here the application of ultrastructure expansion microscopy (U-ExM) to Trypanosoma cruzi. This super-resolution technique increases the spatial resolution of a cell or tissue for microscopic imaging by physically expanding a sample with off-the-shelf chemicals and common lab equipment. As the complete T. cruzi life cycle can be recapitulated in the lab using different cell culture techniques, we describe a detailed protocol for the application of U-ExM in all life cycle stages in vitro.
