This chapter presents protocols for the purification and analytical characterization of major dietary carotenoids such as lycopene, β-carotene, lutein, zeaxanthin, and β-cryptoxanthin from plant and microbial sources. Methods include flash chromatography, column chromatography, and TLC for purification, followed by TLC, UV-Visible spectrophotometry, and HPLC for compound identification and quantification. Purification methods include column chromatography, flash chromatography, and TLC. Characterization is performed using TLC, UV-Visible spectrophotometry, and HPLC. Emphasis is placed on minimizing pigment degradation and ensuring analytical suitability for research and quality control.
