Characterization of Transgenic Rice Plants by Non-radioactive Southern Hybridization

Southern hybridization is a method for detection of specific DNA sequences and gene analysis. It is performed by electrophoresis of the DNA samples after digesting with an appropriate restriction enzyme and transferring it to a nylon membrane and hybridizing it with a labeled probe. In this chapter we describe the protocol for transgene detection in rice plants by capillary transfer of DNA to a nylon membrane and hybridizing it with a Digoxin-labeled oligonucleotide probe and detection using alkaline phosphatase conjugated anti DIG antibody.
