Nanopore sequencing applications allow for the rapid acquisition of genetic information. However, the fast and cost-efficient extraction of high molecular-mass genomic DNA from Komagataella phaffii can become a bottleneck in setting up an effective sequencing pipeline. In this chapter, we describe two methods for the extraction of genomic DNA that are suitable for downstream PCR applications and for long-read genomic DNA sequencing via a Nanopore sequencing device, respectively.
