Cryopreservation is a vital technique for conserving biological material at −196 °C in liquid nitrogen. In this study, we explore the cryopreservation of pollen from wild mango accessions (Appemidi), a unique group known for their distinctive aromatic properties, collected from various locations in the Western Ghats. Ensuring successful cryostorage requires meticulous handling and precise timing, particularly in the collection of freshly dehisced pollen from the field. Both dehisced and nearly dehisced anthers were collected between 10:30 and 11:30 A.M. In vitro pollen germination assays revealed diverse germination patterns. The pollen, after dehydration, was carefully packed in aluminum pouches, stacked in canisters, and gradually lowered into liquid nitrogen for long-term preservation. This chapter outlines the methodology for cryopreserving Appemidi mango pollen, providing insights into its potential applications in conservation and breeding programs.
