Fast and Accurate Enrichment of Canid Excrement for Species* and Other Analyses (FAECES*) Method for Noninvasive Genotyping of Field-Collected Canid Scat
摘要
The FAECES protocol details a method for obtaining multi-locus SNP genotypes from noninvasively collected scat samples. It employs a capture enrichment technique that can be adapted for various vertebrate species. The workflow encompasses probe design using BaitsTools, DNA extraction using the Qiagen DNeasy PowerSoil Pro kit, genomic DNA library preparation via the Santa Cruz Reaction (SCR) protocol, in-solution hybridization capture with myBaits probes, and comprehensive bioinformatic analysis utilizing tools such as FastQC, TrimGalore, Nextflow, and SNPRelate. The protocol aims to differentiate between species, identify individuals, and determine sex from scat samples, emphasizing flexibility and adaptability for diverse research applications.
