The extraction of chloroplastic and chromoplastic pigments (carotenoids and chlorophylls) is one of the most important steps in the analysis of these compounds, and usually consumes significant amounts of solvents and time, as well as requiring well-trained personnel skilled in the handling of these labile compounds. This chapter describes a simple, rapid, and reproducible protocol based on the application of the mechanical-grinding solvent-assisted extraction (MGSAE) technique for the analysis of carotenoids and chlorophylls from plant tissues and plant-derived food prior to their spectrometric and/or chromatographic analysis. Using this protocol, which involves the use of an oscillating ball mill, a pigment extract can be prepared and ready for instrumental analysis in a matter of minutes, with very low extraction solvent consumption (a few milliliters) and minimal sample handling. The amount of sample required ranges from 0.05 to 1.0 g in most cases, and the use of specific adapters allows up to 20 samples to be processed simultaneously.
