Solid-phase peptide synthesis (SPPS) is often the method of choice for the assembly of complex peptides, peptide mimetics, and other biomacromolecules of therapeutic value. Nevertheless, this method is not sustainable and eco-friendly as it requires huge amounts of hazardous organic solvents and reagents. Herein, we describe a method for the assembly of peptides in water or aqueous mixtures on a novel polymeric solid support, the Li resin. Based on polyacrylamide fibers with 50 mesh particle size, it exhibits excellent swelling properties in aqueous media. Moreover, it shows high mechanical stability as well as high loading of functional amino groups (loading capacity 0.5 mmol/g).
