Purification of peptides produced by chemical synthesis or semi-synthesis is required nearly for all sequences. Reversed-phase high pressure (performance) liquid chromatography (RP-HPLC) has long been the purification method of choice for these molecules. But as solid phase synthesis strategies have improved and demand for increasing numbers of peptides continues, alternative purification options are now being developed and employed in the traditional peptide synthesis workflow. Herein we introduce reversed-phase high performance flash chromatography (RP-HPFC) and describe strategies enabling its use as a sole or complimentary purification strategy for traditional RP-HPLC methodologies.
