Loop-Mediated Isothermal Amplification (LAMP)-based diagnostic test can identify Staphylococcus aureus (S. aureus) utilizing two target genes, femA and arcC. The optimal reaction temperature for the femA and arcC genes was determined to be 65 °C and 60 °C, respectively. The technique specifically amplifies DNA from S. aureus only, without cross-reactivity with other similar bacterial species, and demonstrated a 100-fold better sensitivity than PCR. Additionally, when tested in both intact and enriched conditions, the LAMP assay detected the pathogen in food samples, particularly meat and dairy products. Thirteen samples were found to be positive for S. aureus, with LAMP revealing a higher number of positive samples than PCR.
