Implementation of Fluorescent Primers in Loop-Mediated Isothermal Multiplex Assays

Loop-mediated amplification (LAMP) is an example of an isothermal nucleic acid amplification technology. LAMP uses template-specific primers to provide sufficient amplified DNA for detection and quantification. LAMP has numerous benefits over PCR including single temperature reaction conditions enabling the use of low-complexity equipment, high sensitivity, robustness to certain inhibitors, and rapid amplification. However, the specificity of LAMP can be problematic despite the number of primers involved, and false-positive results can occur from nonspecific primer interactions. Detection and quantification methods that record increasing total DNA are therefore inferior to methods that track specifically the LAMP reaction such as the quenched fluorescent primer technique. In this chapter, assay design considerations for LAMP quenched fluorescent primers are described and a multiplexed method for testing with multiple fluorophores is shown.
