Molecular DNA-based methods, such as PCR/qPCR, have been employed to detect yeasts and molds in various food products, offering faster results due to their heightened sensitivity and specificity, compared to culture-based techniques. However, recently, isothermal amplification techniques like loop-mediated isothermal amplification (LAMP) have emerged, simplifying assays and reducing costs. In this chapter a panfungal LAMP assay (LAMP-18S) for the rapid detection of spoilage responsible fungi in fruit preparations is described. Primers targeting the 18S rRNA gene were designed for this purpose. The study explored two distinct detection strategies: real-time fluorescence and naked-eye color detection.
