Determination of Protein Concentration: Sedmak and Grossberg Method

The determination of protein concentration is essential in many biological and biochemical experiments. Various colorimetric methods exist to estimate protein concentration, most of which rely on the formation of color complexes with proteins. The intensity of the color generated is directly proportional to the concentration of proteins in the solution. By measuring the absorbance (Abs) of known protein standards, a calibration curve can be created. This curve correlates the absorbance values of standards with their protein concentrations. Using Beer’s law (Abs = abc) and the calibration curve, the concentration of protein in an unknown sample can be determined. In the Sedmak and Grossberg (SG) method, Coomassie Blue G-250 is used as a color reagent. The reagent forms blue complexes with proteins, which can be quantified by measuring absorbance at two specific wavelengths: 620 nm (protein-bound Coomassie Blue) and 465 nm (free dye).
