Limited research has been published on the characterization and therapeutic potential of human mesenchymal stromal cells (MSCs) derived from hair follicle dermal sheath (DS). Here, we present protocols for isolating and culturing human DS-MSCs, beginning with the use of a dissecting microscope to separate dermal sheaths from hair follicles for trypsin digestion. Additionally, we outline protocols for adipogenic, osteogenic and chondrogenic differentiation of DS-MSCs, as we explore their potential applications in stem cell therapy and tissue engineering.
