Of the modified nucleic acid-based detection methods, a newer technique is developed by the combination of serological assay with molecular assay via ELISA and PCR methods. It provides two-way specificity and double sensitivity, thus making it a robust detection method. In the first part, ELISA is performed using specific antibodies and the captured antigen is then subjected to PCR detection using specific primers in the second part of the protocol. This doesn’t require the extraction of viral RNA unlike conventional PCR, thus referred as Immuno-PCR or Immunocapture PCR. This method stands well on 3 “S”s of the efficiency of a diagnostic method, i.e., sensitivity, specificity, and speed. However, the cost of the setup is still high, hindering its use in day-to-day laboratory diagnostics.
