Application of Digital PCR (dPCR) in Plant Virus Detection

Pathogens present a significant risk to both food safety and public health. Traditional pathogen detection methods are increasingly inadequate for meeting the demands of modern diagnostics and control. Digital PCR (dPCR) has gained substantial devotion due to its capability to precisely quantify pathogens, offering advantages such as high selectivity, ease of use, accuracy, and speed. Droplet digital PCR (ddPCR) is a highly sensitive molecular biology tool utilized to quantify nucleic acids with absolute precision. It fractionates a PCR mixture into thousands of nanoliter-sized droplets using water-oil emulsion technique and each droplet acts as a separate individual micro-reaction during PCR, thus increasing amplification efficiency manifold. It follows a simple workflow wherein portioning is the primary step, followed by PCR amplification in a compatible thermal cycler and finally detection and quantification using software. Compared to traditional PCR methods, it provides increased sensitivity, reduced variability, and enhanced accuracy.
