Detection of Enterotoxigenic Staphylococcus aureus by Nested PCR

Staphylococcal food poisoning is an intoxication that results from the consumption of improperly prepared or stored foods containing sufficient amounts of one or more preformed S. aureus enterotoxins. Therefore, this study was aimed to determine the existence of enterotoxigenic Staphylococcus aureus using nested PCR. A two-step PCR procedure for detection of genes encoding Staphylococcus aureus thermonuclease (nuc) was accomplished using the primers nuc_ext-f (5′ GCGATTGATGGTGATACGGTT 3′) and nuc_ext_int-r (5′GCCAAGCCTTGACGAACTAAAGC 3′) in the first PCR. In the second step, aliquots are analyzed by nested PCR using the following sets of primers nuc_i-f (5′AAAATGCAAAGAAAATTGAAGTC 3′) used in combination with the same primer nuc_ext_int-r of the first PCR (5′ GCCAAGCCTTGACGAACTAAAGC 3′).
