As one of the most basic structural parameters of proteins, molecular size distribution has an important effect on the properties of protein solutions, the processing properties, and the usability of protein. There are many methods for determining the molecular weight of proteins, such as size exclusion-high-performance liquid chromatography (SEC-HPLC), osmotic pressure, light scattering, ultracentrifugation, and polyacrylamide gel electrophoresis. Among these techniques, the SEC-HPLC technique has the advantages of faster, more accurate, better repeatability and automation. Therefore, this chapter provides a comprehensive guidance to the analysis of the molecular size distribution of proteins using SEC-HPLC.
