Isolation, Cryopreservation, and Differentiation Methods of DPSC

A simple and efficient method for isolating the most basic stem cell populations from accessible tissues is essential for tissue engineering. Autologous pulp stem/progenitor cells obtained from deciduous or permanent teeth are a promising source for dental tissue engineering. These cells are easily isolated, readily expandable in vitro, and ethically unobjectionable, making them an attractive model system for various research applications. While the absence of universally recognized markers remains a significant challenge in stem cell isolation, there is also no standardized method for isolating or purifying DPSCs. This chapter explores various methods for isolating DPSCs, examining the impact of different isolation techniques and culture media on stem cell properties. Cryopreservation and banking of DPSCs are also discussed. The chapter delves into tri-lineage differentiation methods and addresses the challenges associated with adipogenic differentiation of DPSCs. Finally, the chapter reviews odontogenic and osteogenic differentiation mechanisms.
