Direct Injection of Microbes into Tissues/Wounds

Injection of microorganisms in plant tissue can be performed using methods like aspersion, infiltration, and direct injection by pricking the stem and by rubbing the leaf surface with an astringent agent. To choose a proper inoculation method will depend on the infectivity nature of the microorganism. In this protocol, we summarize the inoculation methods of foliar aspersion for bacteria, infiltration using direct infiltration in leaves and infiltration with negative pressure in leaf disks, pricking axillary buds with needles or toothpicks for vascular colonizing bacteria, and leaf rubbing for RNA viruses.
