Bacterial tagging with genes encoding for fluorescent proteins is a useful tool to visualize the plant colonization by administered plant growth promotion bacteria using fluorescence microscopy. A stable genetic transformation of the selected strains is preferable for the assessment of their colonization and establishment in the soil or in the plant tissues during long-term, in vivo plant-microbiome engineering experiments. In this chapter, we describe the procedure of conjugation for stable bacteria transformation by using the mini-Tn7 transposon system, an efficient way to genetically manipulate a broad range of Gram-negative bacteria that allows the tagging with a single-copy, stable insertion of the marker gene in the genome.
