Quantification of Metagenomic DNA

To determine the concentration of DNA, one may measure the absorbance at a wavelength of 260 nm (A260) using a spectrophotometer and a quartz cuvette. An absorbance value of 1 unit at a wavelength of 260 nm indicates a concentration of 50 μg of genomic DNA per mL. This assumes a standard path length of 1 cm and a low-salt buffer, such as Tris–HCl with a pH of 7.0.
