摘要

The regeneration of protocorm-like bodies (PLBs) from callus culture is an effective method for clonal propagation and genetic transformation. It offers a useful experimental system to explore the complex patterns and characteristics of morphogenesis in orchids. This chapter details a general protocol for callus induction and subsequent maintenance of callus cultures and regenerated plantlets for Phalaenopsis. The Notes section addresses the application of plant growth regulators during callus induction and maintenance of callus culture. Various carbohydrate sources, including sucrose, maltose, and trehalose, influence the formation and development of PLB from the Phalaenopsis embryogenic callus.
