Mapping Neuron–Astrocyte Circuits with CaMPARIGFAP

CaMPARIGFAP, a calcium-modulated photoactivatable sensor, is a novel optical tool implemented for astrocytes that allows real-time monitoring of astrocytic calcium dynamics in parallel with targeting Ca2+ activity in large areas of tissue with temporal precision. In this chapter, we provide detailed guidelines to map neuron–astrocyte circuits in entire brain regions by combining CaMPARIGFAP with opsin-based protocols to activate specific neuronal pathways, establishing a methodology to expand our understanding of astrocyte involvement in complex circuits.
