Enumeration of Testate Amoeba Through Direct Count from Soil

Several methods have been used to extract testate amoebae from various ecological niche to examine and quantify their tests using light microscopy. Testate amoebae are generally enumerated by direct microscopy from soil slurry which is pre-treated to highlight tests. We here described two methods for enumerating testate amoebae from soil samples; one in which the sample is pre-treated with chemical and other in which sample is sieved and stained to distinguish tests from soil particles.
